Even if you mount your C: drive properly into a different drive letter, chances are your application requires some special libraries or registry keys that it installed while installing in Windows. Naturally Wine doesn't have these, so you'd have to reinstall the application in Wine anyway (unless you know what's missing and you can add it into the wineprefix).
