How to mount the iso is not a Wine question. Ask your distro. 

Once mounted, map the iso as a cd-rom in winecfg (on the drives tab). Note that mounting an iso is not cd emulation, and some apps will not work that way. Search the forum for more information; the issue has come up many times.
