A wineprefix is a directory where Wine's configuration settings and fake C: drive are stored. The default is ~/.wine; that's what you need to delete. A new one will automatically be created the first time you run anything in Wine.

So do this:

Delete ~/.wine
Install the winetricks specified in the AppDB entry
Run the installer for your game

You should be doing this in the latest development release (1.3.12).

If you still have problems, post terminal output, including the command you used to start the program.
