I doubt that. Wine should be capable to pass simple media checks even with
mounted .iso files.
You most likely have some sort of permission problem.
Ok, then lets walk though this step by step.

Please give me:

WINEDEBUG=+tid,+seh,+relay,+volume wine WSC2009.exe &> log.txt 

with the original, _unpatched_ executable and attach to this bug (if too large,
compress it).
Additionally provide info about your iso mount point and block device
permissions:

$ mount | grep iso
$ sudo losetup -a
and then for each listed block device from previous command:
$ ls -l /dev/loopX (X=0,...)
