I don't see any difference with __GL_THREADED_OPTIMIZATIONS=1 and adding
LD_PRELOAD="libpthread.so.0 libGL.so.1" makes the game horrible laggy. All with
the latest nvidia beta drivers (Gforece GTX 275 and i7 quadcore CPU @ 3.6GHz).
My framerate is between 15 and 25 most of the time.

Just to make sure. You are not using ae's altered files, right?
