With the Developer who packages the Windows All in one installer saying:

Recent versions of Gramps AIO (>=5.1.1) are using a slightly patched version of
fontconfig. Fontconfig was having problems on some Windows machines in scanning
for fonts. When it failed, it would be unable to display fonts. Usually a
reboot and reinstall of Gramps would fix it. The newer AIO tries to fix this by
making a more robust update of the fontconfig cache. It seems to be working
better and I have been getting no recent complaints.
