Serial ports work correctly under Wine 1.3.4 - I have six installed (2
on the mobo and four on a six-port PCI serial card). They show up
as /dev/ttyS0 thru /dev/ttyS5. All are visible to a data download
program I run under Wine.

Bear in mind that by default all serial ports are only accessible by
root. If you want to use them from another user, as you will for a Wine
application, you need to change their access permissions. This link says
what I've found out about configuring serial ports for use from normal
users and with Wine applications:
