[Wine] Running wine to an existing windows partition

I use to run double boot system windows/puppy-linux 
C: windows system 
D: data
Puppy linux + swap 

Data are usable by both windows or linux (preferably)

I like to set up wine in order to works directly with the C: drive in order to 
1) save double space / installations 
2) works on most the existing Windows programs 
3) fixe around some wine limitations

Please let me know possibility, exprerience, advises
